Action of plant proteinase inhibitors on enzymes of physiopathological importance Anais da ABC (AABC)

Obtained from leguminous seeds, various plant proteins inhibit animal proteinases, including human, and can be considered for the development of compounds with biological activity. Inhibitors from the Bowman-Birk and plant Kunitz-type family have been characterized by proteinase specificity, primary structure and reactive site. Our group mostly studies the genus Bauhinia, mainly the species bauhinioides, rufa, ungulata and variegata. In some species, more than one inhibitor was characterized, exhibiting different properties. Although proteins from this group share high structural similarity, they present differences in proteinase inhibition, explored in studies using diverse biological models.

Activities of selected digestive enzymes during larval development of large yellow croaker (Pseudosciaena crocea) ArchiMer

Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day I post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. Adaptation of digestive enzymes to dietary protein, carbohydrate and fibre levels and influence of protein and carbohydrate quality in Penaeus vannamei larvae (Crustacea, Decapoda) ArchiMer

Activity of digestive enzymes (trypsin, chymotrypsin and amylase) was studied in relation with food in Penaeus vannamei larvae first stages. Microparticles containing casein involved the decrease of soluble protein content and enzyme activities. Specific trypsin activity but not chymotrypsin was correlated (p<1%) to casein level between 10 and 60% in microparticles. Cellulose fibres in food seemed to have little effect on digestive enzymes. Starch, between 1 and 20% in microparticles, had no influence on specific amylase activity. The effect of different feeding conditions were also tested on growth. Abstract: Candida haemulonii complex (C. haemulonii, C. haemulonii var. vulnera and C. duobushaemulonii) consists of emergent multidrug-resistant pathogens that cause bloodstream and deep-seated infections. However, little is known about their virulence factors. Herein, we evaluated the presence of extracellular serine peptidases in this fungal complex. Serine peptidase activity was measured by spectrophotometry using chromogenic peptide substrates to the S1 family. Chymotrypsin-, trypsin- and elastase-like activities were detected in all fungal isolates. Detection of trypsin inhibitor in seeds of Eucalyptus urophylla and its influence on the in vitro growth of the fungi Pisolithus tinctorius and Rhizoctonia solani BJM

Inhibitors of plant proteases can regulate the hydrolysis of proteins inside the cells and also participate in the mechanisms of plant defense against herbivore insects and pathogens. Here, we demonstrated that seeds of Eucalyptus urophylla exhibit activities of trypsin and papain inhibitors, two proteases commonly found in living cells. Low amounts of proteins of the crude protein extract of seeds and fractions partially purified by gel filtration, with inhibitory activity against trypsin, inhibited in vitro the mycelial growth of a compatible isolate of the ectomycorrhizal fungus Pisolithus tinctorius and allowed an unsatisfactory growth of another isolate from Pinus taeda, considered incompatible for this eucalyptus species. A fisheries research cruise conducted in 2000 offered a first opportunity to take simultaneous measurements of the activities of three enzymes in mesozooplankton samples collected at a regional scale over the continental shelf of the Bay of Biscay in the NE Atlantic, with the aim of characterizing main aspects of the functioning of the biotic environment of small pelagic fish populations. ABSTRACT: The aim of this study was to verify the activity of some digestive enzymes in four fish species with different feeding habits. Knowledge of these enzymatic activities can help us to understand the species' digestive processes. The species chosen for this study were Ctenopharyngodon idella (herbivore), Rhamdia quelen (omnivore), Leporinus obtusidens (omnivore) and Hoplias malabaricus (carnivore). The digestive tract of these species was divided into four portions to estimate enzymatic activity: stomach, anterior, mid and posterior intestine. Glyphosate on digestive enzymes activity in piava (Leporinus obtusidens) Ciência Rural

The effects of glyphosate, a nonselective herbicide (1.0 or 5.0mg L-1) on digestive enzymes activity (stomach and intestine) were evaluated in juveniles of piava (Leporinus obtusidens) after 90 days of exposure. The activity of acid protease, trypsin, chymotrypsin and amylase increased with the increase of glyphosate concentration. These results indicate that glyphosate affects digestive enzyme activities in this species, and may be an indicator of poor nutrient availability when fish survive in herbicide-contaminated water.

Influence of dietary live yeast on European sea bass (Dicentrarchus labrax) larval development ArchiMer

Recent results have demonstrated that live yeast in diets improved gut maturation in sea bass larvae. In this study, we tested the absence and two levels of live yeast in sea bass larvae diets. Specimens were fed from time of mouth opening to 37 days after hatching a diet of 0%, 1.1%, or 5.7% wet weight of live yeast (Debaryomyces hansenii CBS 8339). Yeast incorporation improved survival 10%, and reduced malformed larvae. In groups fed 1.1% yeast, only 1% of larvae were malformed, compared to 14% in the control group. Final mean weight in groups fed 1.1% yeast was twice that of other groups. Purification and characterisation of trypsin-like enzyme from the pyloric caeca of cod (Gadus morhua) II BABT

A trypsin -like enzyme from the pyloric caeca of cod (Gadus morhua) was purified by affinity chromatography on CHOM Sepharose 4B. Some characteristics were established by its catalytic activity on T.A.M.E., typical enzyme substrate, and serine protease inhibitors. The enzyme had an isoelectric point of 5.30 and 5.89 and was very similar in amino acid composition to bovine trypsin, but differed in having a higher relative amount of acidic amino acids and a lower amount of basic amino acids. Reactivity of anti-thyroid antibodies to thyroglobulin tryptic fragments: comparison of autoimmune and non-autoimmune thyroid diseases BJMBR

Studies concerning the antigenicity of thyroglobulin fragments allow the characterization of the epitopes but do not consider the role of heavier antigenic fragments that could result in vivo from the action of endoproteases. Here we assess the relative importance of the fragments obtained from thyroglobulin by limited proteolysis with trypsin and compare by immunoblotting their reactivity to serum from patients with autoimmune (Graves' disease and Hashimoto's thyroiditis) and non-autoimmune (subacute thyroiditis) disease. The results showed no difference in frequency of recognition of any peptide by sera from patients with autoimmune thyroiditis. The role of pancreatic proteolytic enzymes in the excystation process of Eimeria tenella oocysts and sporocysts was studied in vitro. Intact sporulated oocysts were preincubated in phosphate buffer, NaCl 0.9% (PBS) added with 0.5% chicken bile extract in a 5% C02 atmosphere for 30 minutes prior to exposure to either 0.25% (w/v) chicken trypsin, chymotrypsin, pancreatic elastase, or a 1% (w/v) crude extract of unsporulated and sporulated oocysts of E. tenella (Expt.1). Serine-proteases are involved in vital processes in virtually all species. They are important targets for researchers studying the relationships between protein structure and activity, for the rational design of new pharmaceuticals. Trypsin was used as a model to assess a possible differential contribution of hydration water to the binding of two synthetic inhibitors.
